一、Product introduction
BenzoNuclease is a genetically engineered endonuclease derived from Serratia marcescens. BenzoNuclease degrades double-stranded, single-stranded, linear, and circular DNA and RNA, and completely degrades nucleic acids into 5 ‘-monophosphate oligonucleotides 3 to 5 bases long. Because it can efficiently degrade any form of DNA and RNA, it is also known as “omnipotent nuclease”.
BenzoNuclease is an effective tool enzyme for the removal of all forms of DNA and RNA from biological products, whether in laboratory research or in industrial-scale production. Through efficient nucleic acid removal, the effect and yield of follow-up experiments and production can be significantly improved, and the performance is superior to other nucleic acid removal methods.

Activity definition: The amount of enzyme used to change the absorption value of △A260 by 1.0 (equivalent to complete digestion of 37 μg salmon essence DNA into oligonucleotides) in a reaction system of 2.625 mL at 37℃ and pH 8.0 is defined as one activity unit (U).
三. Use method
- Cell treatment: a. Adhesion cell removal media, washed with PBS, 1mLRIPA lysate (or other mammalian cell lysate) added 1-2UL (0.1-1μl) nuclease, incubated at room temperature or on ice for 5-30min, collected lysate, centrifuged supernatant can be carried out downstream experiments. b. After the suspension cells were collected by centrifugation, 1mLRIPA lysate (or other mammalian cell lysate) was added to the centrifuge tube with 1-2uL (0.1-1μl) nuclease, incubated at room temperature or on ice for 5-30min, the lysate was collected, and the supernatant could be centrifuged for downstream experiments.
- Tissue sample: After 30-100mg of animal or plant tissue is fully ground, 100-200uL lysate is added, 0.5-1uL nuclease is added at the same time, incubated at room temperature or on ice for 5-30min, the lysate is collected, and the supernatant can be centrifuge for downstream experiments.
- Escherichia coli or other bacteria: After the bacteria are collected by centrifugation, cracked with lysate or crushed, 0.5-1uL nuclease is added per 1mL, incubated at room temperature for 5-30min, the lysate is collected, and the supernatant can be centrifuged for downstream experiments.
Note: If the solution is high salt (>0.3MNaClass) solution, acidic or alkaline, containing high concentration of detergent and denaturing agent, the enzyme amount or incubation time should be appropriately increased.

The best condition is that more than 90% of the enzyme activity is preserved. The effective condition is that more than 15% of the enzyme activity is preserved.

Storage stability: -20℃ or below storage, two years effective.
Transport stability: refrigerated transport, stable activity.
Note: This product is not recommended to store -800C, repeated freezing and thawing will reduce the enzyme activity of this product.
